UConn Researcher Develops Successful Zika Virus Vaccine in Preclinical Studies
UConn researcher Paulo Verardi, associate professor of pathobiology and veterinary science in the College of Agriculture, Health and Natural Resources, has demonstrated the success of a vaccine against Zika virus and recently published his findings in Scientific Reports, a Nature Research publication. He has also filed provisional patents with UConn’s Technology Commercialization Services for the novel vaccine platform technology used to generate the vaccine, as well as genetic modifications made to the vaccine that significantly enhance expression of the vaccine antigen. Verardi, a Brazilian native, was in Brazil visiting family in the summer of 2015 when the Zika outbreak first began to make waves and soon reached epidemic status. Back in the United States, Verardi kept tabs on the Zika epidemic and its emerging connection to microcephaly, a serious birth defect that causes babies to be born with small heads and underdeveloped brains. In October of that year, Verardi called then-Ph.D.-student Brittany Jasperse (CAHNR ‘19) into his office and told her he wanted to apply their newly developed vaccine platform and start developing a vaccine for Zika virus. Kids in School: 3 Feet Apart or 6 Feet?
The Centers for Disease Control and Prevention (CDC) recently published updated guidelines for elementary schools, "now [recommending] that, with universal masking, students should maintain a distance of at least 3 feet in classroom settings" as opposed to the previous recommendation of 6 feet. While many have had strong reactions for and against this updated guidance, immunologist Joseph Comber, PhD, an associate teaching professor of biology at Villanova University, wants to add nuance to the conversation. On the one hand, Dr. Comber notes, when the CDC updates health and safety guidelines, the public can trust that as more information and data become available, the science behind the updates becomes more accurate. He stresses that studies "showed no difference in transmission between kids kept 3 feet apart or 6 feet apart in schools." Plus, Comber says, "we already know that kids younger than 10 and 11 are less likely to spread the virus than older kids and adults." However, Dr. Comber cautions that the subjects of the study followed "strict mask guidance and adherence to masking. We don't know what happens if schools enforce 3 feet of distance but are not strict with masking." He also points out that where the study was done can have an impact on the results. Comber emphasizes that "community spread is most important to the specific community and, as we've seen so far, different states and regions can spike with infections at different times." He wonders "if this is especially true in areas that have higher virus circulation than others." Dr. Comber notes that these guidelines are for elementary schools only, with one rather large caveat: "children are increasingly being infected." Another concern with these shifting regulations is whether they will impact public trust. Villanova professor Jie Xu, PhD, is an expert on science and health communications. Her concern about this change is minimal "because science is evolving." She emphasizes that "we need to be able to revise conclusions when new evidence comes in," and she sees the country's "highly politicized environment" as something to be taken more seriously, when it comes to public health. Dr. Xu cites "rising distrust in authorities and institutions (which has been going on for quite some time)" and notes that, when it comes to the nation’s children, in-person versus virtual schooling is a "highly charged issue." In the end, according to Dr. Comber, "it remains to be seen what happens when kids are packed 3 feet into classrooms on a broad scale."

What Can A Forgotten Piece of Our Opioid Addiction and Treatment History Teach Us?
As the nation struggles with the third wave of a continuing opioid epidemic, a newly republished book co-authored by Nancy Campbell, the head of the Department of Science and Technology Studies at Rensselaer Polytechnic Institute, offers insight into present-day drug addiction and treatment by exploring a complex chapter from the nation’s past. Written with JP Olsen and Luke Walden, The Narcotic Farm: The Rise and Fall of America’s First Prison for Drug Addicts details the history of the United States Narcotic Farm, a federal institution that opened in 1935 outside of Lexington, Kentucky. Jointly operated from 1935 to 1975 by the U.S. Public Health Service and the Federal Bureau of Prisons, the Narcotic Farm was a combination prison, hospital, working farm, rehabilitation center, and research laboratory. “All of our scientific knowledge about human opioid addiction comes from that time, comes from that place,” said Campbell, a leading figure in the social history of drugs, drug policy, and harm reduction, on an episode of the Landmark Recovery Radio podcast. The facility, which was also the subject of a 2009 documentary featuring Campbell, has a complicated legacy. It revolutionized treatment methods commonly accepted today, such as using methadone to medically manage heroin detox and the development of drugs like naloxone and buprenorphine. But it fell under a cloud of suspicion in 1975, when Congress learned that researchers had recruited patients as test subjects for CIA-funded LSD experiments as part of the notorious MK-Ultra project. “With the ongoing opioid epidemic worsening this past year in the midst of the COVID-19 pandemic, the lessons learned in this book continue to be relevant today,” Campbell said. Campbell is also the author of Using Women: Gender, Drug Policy, and Social Justice and Discovering Addiction: The Science and Politics of Substance Abuse Research, as well as the co-author of Gendering Addiction: The Politics of Drug Treatment in a Neurochemical World. Her most recent book, OD: Naloxone and the Politics of Overdose, was published in 2020. “Nobody should die of overdose. A high overdose death rate signals that we have not cared for the people who have been hurt most by the war on drugs, first pursued by President Dwight D. Childhood psychology expert on migrant children at U.S.-Mexico border
Kathryn Humphreys, assistant professor, department of psychology and human development, is available for commentary on the influx of migrant children at the border and group-based housing for these children. Kathryn received her doctoral degree in clinical psychology and has expertise in infant and early childhood mental health. Her research focuses on characterizing the early environment and examining links to later life outcomes. Some of her recent research finds that stressful or traumatic experiences occurring in a child’s earliest years—birth to age 5—have been linked to reduced hippocampal volume in adolescence, which is connected to learning, memory and mood.

Evaluating the Impact of Facebook's Ban on Vaccine Misinformation
A new Facebook policy has banned misinformation about all vaccines on its platform. Villanova University professor Jie Xu, PhD, who specializes in science and health communication, examined this decision. "On one hand, there clearly is a lot of mis/disinformation on social media regarding vaccines; some of them are simply uninformed and, of course, harmful to public health," said Dr. Xu. "On the other hand, many details relating to the COVID-19 vaccine, in my view, are still open to scientific debate." So, what determines what is labelled misinformation? Dr. Xu believes this is a complicated determination. "Science itself is evolving with falsification and revision to previous claims when new evidence comes in," Dr. Xu noted. "Who is to say that some claims deemed true at this moment won't be overturned in the future? What are the standards to be used in defining what is true information or misinformation? And perhaps more importantly, who are the 'fact-checkers' that are considered trustworthy to the majority of Americans?" However, there are some benefits to Facebook's decision. "On a more positive note, there is some preliminary evidence indicating that labeling misinformation on social media may help to alleviate the negative influence of vaccine misinformation claims," Dr. Xu said. "The challenge is that the people that are most susceptible to misinformation, and those that health professionals really want to reach out to, are the ones that have the least level of trust on this type of intervention. In some corners, this will likely to be viewed as violation to free speech and perhaps backfire." How does Facebook's banning align with free speech? "My understanding of free speech is that it's not that we don't pay a price for it—unless it's inciting violence, most information has been allowed to flow relatively freely—but it's that the alternative could be much worse," said Dr. Xu. "At the end of the day, we need to create an environment in which honest, open and critical conversations are welcomed, and we do need each other to find the truth."

Ask an Expert: What is COVID-19’s impact on the homelessness crisis?
The COVID-19 pandemic continues to impact the homeless community and homelessness crisis, including posing unique health risks to the homeless population and spurring a likely increase in homelessness due to job losses. “People experiencing homelessness are at enormous risk of exposure to the coronavirus, due to inability to self-isolate, as evidenced by outbreaks in congregate shelters,” says Marybeth Shinn, Cornelius Vanderbilt Chair and professor of human, organizational and community development at Vanderbilt Peabody College of education and human development. “With the cold weather coming, service providers are scrambling to provide food, shelter and outreach services safely, and to use rental assistance to get people into housing.” Shinn also explains that while eviction moratoriums imposed during the pandemic work to delay evictions, they do not prevent them. Arrears for rent, utilities and fees continue to accumulate when the moratorium ends, and landlords can continue to charge late fees for late payments. On the one hand, moratoriums will help keep many renters in their homes at a time when the alternatives, such as crowding in with friends and relatives or even becoming homeless, puts people’s health at risk. At the same time, landlords, especially small landlords, are also suffering. Landlords often have mortgages as well as other expenses to pay, relying on rental income to do so. In her new book with Abt Associates researcher Jill Khadduri, In the Midst of Plenty: Homelessness and What to Do About It, Shinn argues that homelessness is not a result of personal failure, but rather societal failure, as we have the knowledge and resources to end homelessness but lack the political will. As an immediate step during the pandemic, Shinn advises that Congress needs to enact relief for tenants and landlords, as well as reinstate weekly supplements to unemployment benefits to help people stay current on rent.

COVID-9 is changing. With variants from places like Britain, Brazil, and South Africa surfacing and presenting in patients around the globe, the virus that scientists are trying to trap seems to be finding ways to wiggle free. The topic has many in the scientific and health care communities concerned, and recently, the New York Times featured leading experts to help explain what is happening and what American’s need to know to stay safe and healthy. Now, Dr. Denison and Sandra Weller, a virologist at the University of Connecticut School of Medicine, are investigating whether this insight could treat people with Covid. Certain antiviral drugs such as remdesivir fight infections by serving as RNA decoys that gum up the viral replication process. But these medications don’t work as well as some had hoped for coronaviruses. One theory is that the nsp14-ExoN enzyme chucks out the errors caused by these drugs, thereby rescuing the virus. Dr. Denison and Dr. Weller, among others, are looking for drugs that would block the activity of nsp14-ExoN, allowing remdesivir and other antivirals to work more effectively. Dr. Weller likens this approach to the cocktail therapies for H.I.V., which combine molecules that act on different aspects of the virus’s replication. “We need combination therapy for coronaviruses,” she said. Dr. Weller notes that nsp14-ExoN is shared across coronaviruses, so a drug that successfully suppresses it could act against more than just SARS-CoV-2. She and Dr. Denison are still at the early stages of drug discovery, testing different molecules in cells.
